In plain terms: Measures the total concentration of protein in a blood sample - overwhelmingly albumin plus the various globulins - run on a chemistry analyzer rather than estimated with a refractometer, and drawn from blood rather than urine or another body fluid. It is a standard line on comprehensive metabolic testing. Taken alone it is a blunt instrument; its usefulness comes from reading it next to albumin, because the gap between the two indicates whether the globulin fraction is raised, as it can be in chronic inflammation, liver disease, or plasma-cell disorders such as myeloma. Think of the standard blood panel drawn at an annual visit, where this is one of a dozen values reported. It becomes interesting when total protein is high but albumin is normal or low, which can be the first clue that prompts a doctor to look further at the immune proteins. You'll typically see CPT 84155 on a bill or explanation of benefits (EOB) when a clinician performs or bills for this service.
