In plain terms: A first-pass urine drug screen read by eye, with no analyzer involved - the dipstick, cup, or test card develops colored lines that a person interprets directly. It can cover any number of drug classes in a single test device, and the whole encounter on one day is reported once regardless of how many substances the device checks for. This is the simplest of the presumptive-screening tiers; sibling codes cover screens read with the help of an instrument reader or run on a chemistry analyzer. A presumptive result only says a drug class is probably present or absent, so anything that matters clinically is usually sent on for confirmatory testing. Think of the plastic cup screen done in the office while you wait, where the staff member reads the result lines a few minutes later and tells you the answer before you leave. You'll typically see CPT 80305 on a bill or explanation of benefits (EOB) when a clinician performs or bills for this service.
